Transcription profiling of whole testis from wild type, bgcn- and Os+bgcn- Drosophila [GSE4188]
Whole testes were dissected from adult males. RNA from five or six biological replicates were generated and the expression profiles were determined using Affymetrix Drosophila Genechip 1 arrays. Comparisons between the bgcn- and Os+bgcn- groups allowed for the identification of stem cell genes.
Transcription profing of Drosophila tissues in adult and larva [GSE7763]
A new online resource (flyatlas.org) provides the most comprehensive view yet of expression in multiple tissues of Drosophila. Meta-analysis of the data reveals that a significant fraction ofthe genome is expressed with great tissue specificity in the adult, demonstrating the need for the functional genomic community to embrace a wider range of functional phenotypes. Well-known developmental genes are often re-used in surprising tissues in the adult, suggesting new functions. The homologues of many human genetic disease loci show selective expression in Drosophila tissues with function analogous to the affected tissues in the cognate human disease, providing a useful filter for potential candidate genes. Experiment Overall Design: FlyAtlas is composed of data covering 8 distinct adult tissues (brain, head, midgut, Malpighian tubules,hindgut, testis, ovary, male accessory glands) so far, together with two larval tissues (tubule, fat body), each run with 4 replicates on Affymetrix Dros Genome 2 chips (with probesets for 18770 transcripts), and compared with a matched whole-fly sample.
Testes dissected from 1-5 day adult y[*] w[67c1]/Y males raised at 25oC. RNA isolated using Trizol (Life Technologies) and a single round of Poly(A)+ selection using Oligotex (Qiagen). cDNA library constructed using Stratagene ZAP-cDNA syntesis kit. Oligo dT-primed, size fractionated ~1-6 kb, and directionally cloned at EcoRI and XhoI in Uni-ZAP XR. Following a single round of amplification pBlueScript SK phagemids were mass excised. A distribution channel for clones is being sought, but not currently available. Requests for clones cannot be honored.

The mRNA for the testis library was made from testes and seminal vessicles hand dissected from 0-3 day old Ore-R males. RNA kindly provided by the lab of Margaret Fuller. Sized fractionated cDNAs were directly ligated into pOTB7. Plasmid cDNA library.
